A Mouse Model for Corneal Neovascularization by Alkali Burn

نویسندگان

چکیده

Corneal neovascularization (CoNV), a pathological form of angiogenesis, involves the growth blood and lymph vessels into avascular cornea from limbus adversely affects transparency vision. Alkali burn is one most common forms ocular trauma that leads to CoNV. In this protocol, CoNV experimentally induced using sodium hydroxide solution in controlled manner ensure reproducibility. The alkali model useful for understanding pathology can be extended study angiogenesis general because avascularity, transparency, accessibility cornea. work, was analyzed by direct examination under dissecting microscope immunostaining flat-mount corneas anti-CD31 mAb. Lymphangiogenesis detected on anti-LYVE-1 edema visualized quantified optical coherence tomography (OCT). summary, will help advance existing assays discover new treatment strategies pathologic extraocular angiogenesis.

Inhibition of mouse alkali burn induced-corneal neovascularization by recombinant adenovirus encoding human vasohibin-1

PURPOSE To evaluate the activity of recombinant adenovirus encoding human vasohibin-1 (Ad-Vasohibin-1) on mouse corneal neovasularization induced by alkali burn. METHODS For the treatment group, 50 mice each received subconjunctival injection (5 microl) of 10(9) plaque forming units of replication-defective Ad-Vasohibin-1. Control group mice received the same dosage of blank adenoviral vector...

Minocycline Inhibits Alkali Burn-Induced Corneal Neovascularization in Mice

The purpose of this study was to investigate the effects of minocycline on alkali burn-induced corneal neovascularization (CNV). A total of 105 mice treated with alkali burns were randomly divided into three groups to receive intraperitoneal injections of either phosphate buffered saline (PBS) or minocycline twice a day (60 mg/kg or 30 mg/kg) for 14 consecutive days. The area of CNV and corneal...

Plasminogen kringle 5 inhibits alkali-burn-induced corneal neovascularization.

PURPOSE Plasminogen kringle 5 (K5) is a potent angiogenic inhibitor. The purpose of the present study was to evaluate the therapeutic effect of K5 on alkali-burn-induced corneal neovascularization (NV) and to investigate its mechanism of action. METHODS Corneal NV was induced in rabbits by NaOH. The rabbits received eye drops containing K5 or vehicle alone, four times per day. Corneal NV and ...

Thalidomide prevents donor corneal graft neovascularization in an alkali burn model of corneal angiogenesis.

OBJECTIVE Thalidomide is a potent inhibitor of angiogenesis. We evaluated the effects of Thalidomide on corneal angiogenesis and on tissue survival of grafts in rabbit eyes with pre-existing neovascularization secondary to alkali burn. METHODS Sixteen rabbits received alkali burns to one cornea. One month post-injury, assessments of corneal neovascularization were performed followed by cornea...

Thalidomide prevents Corneal Angiogenesis in an Alkali Burn model of Rabbit Corneal Neovascularization

Objectives: To determine if Thalidomide has a similar effect in an alkali burn model of corneal neovascularization, we evaluated the effect of Thalidomide on corneal angiogenesis after alkali corneal burns in rabbits. Methods: Twelve rabbits received alkali burns to one cornea. Post injury, four rabbits received oral Thalidomide, 200 mg/kg/day and eight rabbits received placebo (powdered sugar)...
